    摘要

    玉米蛋白/短肽超分子水下黏合剂的构建与生物应用 聂俊莲,文光,李文* 吉林大学超分子结构与材料国家重点实验室,吉林省长春市前进大街2699号,130012 *Email: wenli@jlu.edu.cn 蛋白质和短肽的组合为创造新型的生物材料开辟了新思路。本研究以天然植物蛋白和短肽为构筑单元通过简单的共组装方法开发了一种新型的生物黏合剂。利用十二烷基硫酸钠对疏水玉米蛋白进行分散形成稳定的蛋白胶束,与阳离子短肽通过多重静电/氢键等超分子相互作形成黏合剂。搭接剪切实验表明该黏合剂具有良好的水下粘接性能。体外和体内实验表明该黏合剂具有良好的抗溶胀性、低细胞毒性、血液相容性和炎症反应。爆破压实验表明该黏合剂可抵抗78 cmH2O以上的压力,足以满足硬膜缺损的密封要求。降解实验表明该黏合剂在体内可以两周内解离和降解,与硬膜修复的时间吻合。综合以上优势将该黏合剂成功应用于硬膜缺损的密封和修复。 关键词:玉米蛋白;短肽;共组装;超分子聚合物;生物黏合剂 参考文献 [1] Nie, J.; Sun, Y.; Chen, X.; Wen, G.; Liu, X.; Cheng, M.; Zhao, J.; Li, W. Here, a new kind of biological adhesive based on a natural occurring plant protein and short peptides via a simple co-assembly route were explored. The hydrophobic zein was supercharged by sodium dodecyl sulfate to form stable protein colloid, which intended to interact with cationic short peptide via multivalent ionic and hydrogen bonds. Lap shear experiments show that the adhesive has strong underwater adhesion property. In vitro and in vivo assays demonstrate that the zein/peptide adhesive show excellent anti-swelling property, low cell cytotoxicity, hemocompatibility and inflammation response. Bursting pressure test suggests that the adhesive can resist over 78 cmH2O of bursting pressure, which was enough to meet the sealing requirements of dural defects. Degradation experiments show that the adhesive could in vivo dissociated and degraded within two weeks, which can well match with the time-window of the dural repairing. All these comprehensive features enable the zein/peptide adhesive to be explored as surgical adhesive for successful sealing and repairing the dural defect.
