天然药物化学及化学生物学
主要业绩
申请人曾获国家优青、云南省云岭学者及科技部重大新药创制等重要项目支持,聚焦西部特色药物资源的天然产物研究:①完成了100余种西部地区药用植物的系统研究,分离并改造获得5000余个化合物,其中新化合物1181个,新骨架50个;②获得新型抗HIV病毒功能分子“二苯胺酯”及靶向PD-1/PD-L1肿瘤免疫功能分子Xci-2等多个候选药物分子;③完成了抗HIV候选药物分子“二苯胺酯”成药性评价以及“益母草注射液”化学物质基础鉴定和质量标准提升;④针对天然产物生物活性的高效率发现及深度挖掘的技术瓶颈问题,创新性地开展了基于人工智能的天然药物化学研究,率先针对炎症性疾病构建了天然产物抗炎免疫活性数据库并开展了化学信息学研究。以第一或通讯作者发表高水平论文105篇。申请人聚焦西部特色药用植物资源,借助人工智能技术等多学科融合交叉,重点研究天然来源的抗病毒及免疫调节先导分子的发现及功能评价。具体成果体现如下:
1. 天然药物资源中新型结构天然小分子的发现为新药研发提供分子模板
围绕天然药物资源中新类型、新结构的天然产物的发现和活性研究,对马鞭草科、 益母草属、五味子科等 100 余种药用植物的化学成分进行了研究,发现了 1181 个新的结构,尤其发现了具有多种骨架类型的 190 余个高氧化度、结构新奇的降三萜类化合物。连续不断地发现了系列新的骨架和新类型的天然分子,并首次提出“Schisandra nortriterpenoids(五味子降三萜)”概念; 从马鞭草科植物黄毛豆腐柴中及从中药泽漆中分离得到系列结构新颖的二萜化合物 (Org. Lett. 2020, doi.org/10.1021/acs.orglett.0c02676; Org. Lett. 2018, 20, 6314–6317)。研究成果在不同领域的渗透影响力广泛,引文涉及 43 个学科,50 个国家及地区,568 多所国内外高等学府和研究机构,主要分布在化学、药理学、植物科学等学科领域。发表的新颖天然产物,由于结构的独特性、 创新性,被天然产物化学丛书《三萜化学》《现代天然产物化学》以及《高速发展的中国化学》等书籍所引用和评价。
2. 基于分子靶标和信号通路的新颖天然产物创新发现及活性评价
天然产物的“结构多样性”是其“生物活性多样性”产生的化学基础,通过对课题组的天然产物分子库针对药物靶标、细胞信号通路、疾病模型等进行研究,取得系列创新研究成果,发现了抗肝癌、抗HIV、抑制NLRP3炎症小体活化的作用、调节蛋白泛素化降解以及雌激素合成调节的功能小分子,部分研究成果在 Journal of Medicinal Chemistry, Org Lett, Sci Rep, J Nat Prod 等杂志上公开发表,获得了专利进行知识产权的保护。
3. 创新性地开展基于人工智能的天然活性分子研究
对如何高效开展天然药物物质基础和功能分子快速发现的方法学进行了创新性探索研究。首次构建了天然产物抗炎活性数据库并开展了系统性的化学信息学研究(J. Chem. Inf. Model. 2019, 59, 66-73)。在天然产物权威评述期刊Nat Prod Rep上发表文章,对基于机器学习的天然产物活性研究的进展进行了综述,并结合本团队的研究经验,提出了一系列研究建议,拟在数据科学家和药物研究者之间架设一道有效沟通的桥梁(Nat. Prod. Rep., 2021, 38, 346-361)。借助人工智能药物发现技术,发现靶向PD-1/PD-L1的抗肿瘤免疫皂苷类化合物并开展了潜在应用评价。
在完成为本科生和研究生授课工作的基础上,重要成果在国际学术期刊 Nat Prod Rep, Org Lett, Chem Eur J, J Nat Prod 等杂志上共发表 206 篇研究结果;以第一和通讯或共同通讯作者共发表成果105 篇,影响因子>5.0 的 17 篇;申报了系列专利对研究成果进行了知识产权的保护。
获得中国化学会青年化学奖、中科院西部学者突出贡献奖、中国药学会施维雅青年药物化学奖、云南省青年科技红河奖、以及中科院卢嘉锡青年人才工作奖等10余项奖励和荣誉。
